GC-MS Determination of Volatile Components of Tobacco Leaves with the Algorithm of Heuristic Evolving Latent Projection
摘 要
应用气相色谱-质谱法及直观推导式演进特征投影(HELP)算法测定了烟叶中的挥发性组分。取经干燥及粉碎的烟叶样品用加速溶剂萃取法在160 ℃用乙醇提取15 min,所得提取液经吹氮蒸缩至1.5 mL,取1 μL进样分析,采用DB-5MS毛细管柱进行分离。质谱分析采用电子轰击离子源,在质荷比(m/z) 30~500范围内进行扫描,用HELP算法将所得二维数据解析为各组分的纯质谱及色谱图,将所得纯质谱图与NIST质谱库中的标准谱图作相似性检索,以此作定性分析。通过总体积积分法和归一化法对各组分作定量分析。在得到解析的78个色谱峰中,62个峰的相应组分得到定性鉴定和定量测定。所测得挥发性组分的总量占挥发油总量的88.51%。
Abstract
GC-MS with the heuristic evolving latant projection (HELP) was applied to the analysis of tobacco leaves for its volatile components. The dried and powdered sample of tobacco leaves was extracted with ethanol at 160 ℃ for 15 min by ASE. The extract was evaporated to 1.5 mL by N2-blowing. One μL was taken for analysis using DB-5MS capillary column in chromatographic separation, EI source and scanning range of m/z 30-500 in MS determination. The bi-dimensional data obtained were resolved into pure mass spectra and chromatograms by the HELP method. Qualification was performed by similarity searching of the pure mass spectrum with the standard spectrum in NIST database, and quantification of the components was carried out by integration of total volume and nor malization. Totally 78 chromatographic peaks were resolved, among which 62 components were identified and determined, amounting to 88.51% of the total amount of volatile oils.
