RP-HPLC Determination of Resveratrol in TCM Polygonum Cuspidatum
摘 要
提出了反相高效液相色谱法测定虎杖中白藜芦醇含量的方法。以ODS-C18色谱柱(200 mm×4.6 mm,5 μm)为分离柱,以乙醇-水(2+3)溶液作流动相淋洗,在紫外光检测波长304 nm处进行测定。白藜芦醇的质量浓度在0.05~0.40 mg·L-1范围内与峰面积呈线性关系,方法的检出限(3S/N)为10 μg·L-1。方法用于中药材虎杖中白藜芦醇含量的测定,标准加入平均回收率为98.5%,测定值的相对标准偏差(n=5)为2.6%。
Abstract
RP-HPLC was applied to the determination of resveratrol in polygonum cuspidatum. The ODS-C18 column (200 mm×4.6 mm, 5 μm) was used as stationary phase, and a mixture of CH3CH2OH and H2O (2+3) was used as mobile phase. The content of resveratrol was determined by UV-detection at 304 nm. Linear relationship between values of peak area and mass concentration of resveratrol was kept in the range of 0.05-0.40 mg·L-1, with detection limit (3S/N) of 10 μg·L-1. The proposed method was used in the determination of resveratrol in polygonum cuspidatum, giving values of average recovery obtained by standard addition method of 98.5% and RSD (n=5) of 2.6%.
