Rapid Spectrophotometric Determination of Residual Amounts of 6 Aminoformate Pesticides
摘 要
基于酶抑制法,提出了分光光度法测定6种氨基甲酸酯类农药残留量的方法。采用正交试验优化了获得乙酰胆碱酯酶最佳活性的条件:① 酶与底物的浓度比为6比1;② 显色剂用量为0.4 mg;③ 缓冲溶液的pH为8.5;④ 反应温度为40 ℃。在优化的试验条件下,6种氨基甲酸酯类农药的质量浓度在一定的范围内与抑制率I呈线性关系。方法用于果蔬样品分析,并在此基础上进行加标回收试验,回收率在82.0%~109%之间,测定值的相对标准偏差(n=6)在2.4%~3.6%之间。
Abstract
Based on enzyme inhibition, a method of spectrophotometric determination of residual amounts of 6 aminoformate pesticides was proposed. By orthogonal test, the optimum conditions to obtain favourable acetylcholinesterase activity were shown as follow: ① ratio of concentration of enzyme to substrate: 6∶1; ② amount of chromogenic agent: 0.4 mg; ③ pH value of buffer solution: 8.5; ④ temperature of reaction: 40 ℃. Linear relationships between values of inhibition rate I and mass concentration of the 6 aminoformate pesticides were kept in the definite ranges. Samples of fruits and vegetables were analyzed by the proposed method and test for recovery was made by standard addition method, giving values of recovery in the range from 82.0% to 109%, with RSD′s (n=6) in the range from 2.4% to 3.6%.
