HPLC Determination of Aristolochic Acid A in Cosmetic
摘 要
提出了高效液相色谱法测定化妆品中马兜铃酸A的含量。化妆品样品用甲醇超声提取15 min后,所得提取液经0.45 μm滤膜过滤。以Eclipse XDB-C18色谱柱为固定相,以甲醇和0.5%(体积分数)乙酸(70+30)混合溶液为流动相,用二极管阵列检测器于波长250 nm处检测。马兜铃酸A的质量浓度在0.6~300 mg·L-1范围内与其峰面积呈线性关系,方法的检出限(3S/N)为0.2 mg·L-1。以化妆品样品为基体进行加标回收试验,所得回收率在90.6%~102%之间,相对标准偏差(n=5)在0.85%~3.4%之间。
Abstract
A method of HPLC for the determination of aristolochic acid A in cosmetics was proposed. The cosmetic sample was extracted twice ultrasonically with methanol for 20 min each, and the extract was filtered on 0.45 μm filtering membrane. In the separation Eclipse XDB-C18 column was used as stationary phase and a mixture of methanol and 0.5% (φ) acetic acid solution (70+30) was used as mobile phase. The eluate obtained was determined with DAD at the wavelength of 250 nm. Linear relationship between values of peak area and mass concentration of aristolochic acid A was kept in the range of 0.6-300 mg·L-1, with detection limit (3S/N) of 0.2 mg·L-1. On the base of cosmetic sample, test for recovery was made by standard addition method; values of recovery found were in the range of 90.6%-102% with RSD′s (n=5) in the range of 0.85%-3.4%.
