Study of Chiral Separation of Pseudoephedrine by High-Performance Capillary Zone Electrophoresis
摘 要
以三甲基-β-环糊精(TM-β-CD)为手性选择剂,用毛细管区带电泳法对手性药物伪麻黄碱进行了分离.系统地考察了不同的手性选择剂及其浓度、缓冲溶液的浓度和pH、分离电压等对分离的影响.结果发现,在Tris-H3PO4(10 mmol·L-1 Tris,5 mmol·L-1 H3PO4,pH 2.74)缓冲溶液中,TM-β-CD为15 mmol·L-1,分离电压18 kV条件下,伪麻黄碱对映体在6 min内获得快速基线分离.由此建立了可靠、快速的分析方法.
Abstract
Using trimethyl-β-cyclodextrin (TM-β-CD) as chiral selective reagent,chiral separation of pseudoephedrine by capillary zone electrophoresis was studied. A thorough and systematic examination of the factors,i.e. different kinds of chiral selective reagent and their concentrations,suitable acidity (pH values) and concentration of buffer solution,separation voltage and etc,on the efficiency of separation,was made. It was found that in a buffer medium of Tris-H3PO4 buffer solution (10 mmol·L-1 Tris-5 mmol·L-1 H3PO4,pH 2.74),under the separation voltage of 18 kV,and with the concentration of TM-β-CD to be 15 mmol·L-1,rac-pseudoephedrine enantiomer was separated successfully from the baseline within 6 min. This new method was proved to be reliable and feasible to be used as a rapid analytical method in the research and preparation of pharmaceuticals in clinical studies.
