Comparison of Rapid Determination of Residual Malachite Green in Crucian by HPLC and Enzyme Immunoassay
摘 要
采用高效液相色谱(HPLC)法与酶免疫法(EIA)两种方法检测鲫鱼中残留显/隐色孔雀石绿的含量,对这两种分析方法所得的回收率进行了比较 ,当添加浓度在5.0~50.0 μg·kg-1时,其中HPLC法所测得的回收率为68.2%~90.0%,相对标准偏差小于7.0%,检出限为2 μg·kg-1;EIA法所测得的回收率为72.4%~92.3%,相对标准偏差小于6.8%,检出限为1 μg·kg-1;两种方法均具有高效、快速、稳定的特点,能满足日常检验工作的需要.
Abstract
High performance liquid chromatography (HPLC) and enzyme immunoassay (EIA) were separately applied to the rapid determination of residual malachite green (MG) and leucomalachite green (LMG) in crucian samples. Values of recovery of both methods found were compared. When the standard of MG and LMG were added in the range from 5.0-50.0 μg·kg-1 to crucian samples,the mean values of recoveries of HPLC determinations ranged from 68.2% to 90.0% with RSD′s (n=7) less than 7.0%,and detection limit of 2 μg·kg-1;while the mean values of recoveries of EIA determinations ranged from 72.4% to 92.3%,with RSD′s (n=7) less than 7.0%,and detection limit of 1 μg·kg-1. Both of the methods were characterized by their high efficiency,rapidity and stability,and were feasible to be used in the daily routine analysis.
