HPLC Determination of Polyphenols in Tea
摘 要
应用高效液相色谱法测定了茶叶,特别是绿茶中的8种多酚,包括儿茶素(C)、表没食子儿茶素(EGC)、没食子儿茶素(GC)、表儿茶素(EC)、表没食子儿茶素没食子酸酯(EGCG)、没食子儿茶素没食子酸酯(GCG)、表儿茶素没食子酸酯(ECG)及儿茶素没食子酸酯(CG).在C18反相柱(250 mm×4.6 mm,5 μm)上进行分离,用不同体积比混合的流动相A及流动相B的混合液作梯度淋洗,流速为1.0 mL·min-1,柱温为40 ℃,在280 nm波长处进行紫外检测.上述8组分在0.004~4.0 g·L-1范围内呈线性关系,方法的检出限在0.6~2.6 mg·L-1范围内,平均回收率为83%.
Abstract
High performance liquid chromatography (HPLC) was applied to the rapid determination of polyphenols,including catechin (C),epigallocatechin (EGC),gallocatechin (GC),epicatechin (EC),epigallocatechin gallate (EGCG),gallocatechin gallate (GCG),epicatechin gallate (ECG) and catechin gallate (CG),in tea,especially in green tea.The polyphenols were separated on the C18 RP-column (250 mm×4.6 mm,5 μm) by gradient elution using mixed solutions of mobile phase A and mobile phase B in different proportions.Mobile phase A was a solution of methanol and formic acid mixed in the ratio of 99.7 to 0.3 by volume,and mobile phase B was a dil.solution of formic acid in water,containg 3 vol.of formic acid in 1 liter of solution.Flow-rate of mobile phase was set as 1.0 mL·min-1,and the column temperature of 40 ℃ wa taken.UV-detection at 280 nm was adopted in the determination.Linearity was kept in the range from 0.004-4.0 g·L-1,with detection limits (S/N=3) in the range from 0.6-2.6 mg·L-1,Value of average recovery found was 83%.
