Determination of the Compositional Monosaccharides of Polysaccharides in Aloe by HPGPC,HPLC and ESI-MS
摘 要
利用高效凝胶渗透色谱(HPGPC)测定芦荟浓缩液和芦荟粉中多糖的纯度和相对分子质量,使用1-苯基-3-甲基-5-吡唑啉酮(PMP)作为柱前衍生试剂分别对样品和其水解产物进行衍生,结合高效液相色谱(HPLC)分离和电喷雾-离子阱质谱(ESI-MS)检测分析其单糖组成.结果表明:组成芦荟中多糖的单糖主要是甘露糖和葡萄糖.
Abstract
High performance gel permeation chromatography (HPGPC) was applied to the determination of purity and relative molecular mass of polysaccharides in concentrate liquor and powder of aloe.The aloe sample (in liquor or powder state) or its hydrolysis product was pre-column derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP),and the compositional monosaccharides were separated and determined by hyphenation of HPLC with electron spray ionization-mass spectroscopy (ESI-MS).It was found that mannose and glucose were the main compositional monosaccharides of polysaccharides in aloe.
