Spectrophotometric Determination of Serotonin Derivatives in Safflower Seeds with Ehrlich Reagent
摘 要
基于血清素衍生物与对-二甲基氨基苯甲醛(又称为埃尔利希试剂)之间的显色反应,提出了用分光光度法测定红花籽提取物中血清素衍生物的总量,对此方法的反应机理及反应的主要影响因素作了研究.结果表明:此显色反应应遵循了吲哚环在2号(或3号)空位上的亲电取代反应的规律,呈色反应产物的吸收峰位于625 nm波长处,在此条件下,溶液的吸光度与血清素衍生物浓度在0.025~0.5 mmol·L-1之间呈线性关系,应用此方法测定了新疆红花籽提取物中血清素衍生物的总量,并在此基础上用标准加入法作方法的回收率及精密度试验,测得回收率在97.5%~101.7%之间,相对标准偏差值(n=7)均小于2%..
Abstract
Based on the color reaction between derivatives of serotonin and p-dimethylaminobenzaldehyde (also named as Ehrlich reagent),total amount of serotonin derivatives in extracts of safflower seeds was determined by spectrophotometry.Main influential factors and reaction mechanism of the method were studied.It was found that the mechanism of the color reaction was in compliance with the electrophilic substitution of the indole ring at its 2nd carbon position.Absorption maximum of the colored complex was found at the wavelength of 625 nm,and linear relationship between values of absorbance and concentration of serotonin derivative was found in the range from 0.025-0.5 mmol·L-1.Tests for recovery and precision of the method were made on the base of several samples of extracts of safflower seeds taken from Xinjiang,giving values of recovery in the range of 97.5%-101.7% and values of RSD′s (n=7) less than 2%.
