Mechanism of Interaction of the Coordination Complex Sm(Ⅲ)(CAI)3 with DNA
摘 要
应用吸收光谱、荧光光谱和粘度法并以吖啶橙作探针研究了Sm(Ⅲ)(CAI)3与DNA的相互作用.研究表明,钙黄绿素(CAI)与钐(Ⅲ)形成了配合物,其结合比n钐(Ⅲ)∶nCAI=1∶3,生成八面体构型稀土配合物,Sm(Ⅲ)(CAI)3与DNA的结合常数K=2.09×104L·mol-1.Scatchard方程表明该配合物与DNA的结合为中等强度的嵌插和部分静电作用.
Abstract
Mechanism of the interaction of the coordination complex of samarium(Ⅲ) and calcein (CAI in short) with DNA was studied by spectrophotometry,fluorophotometry and viscositometry using acridine orange (AO in short) as probe.As shown by experimental results,the coordination complex formed between Sm(Ⅲ) and CAI has an octahedronal molecular structure with its binding mole ratio (nSm(Ⅲ)∶nCAI) of 1 to 3,and the Sm(Ⅲ)-CAI complex reacts with DNA to form an association complex with the combination constant (K) of 2.09×104L·mol-1.As shown by ploting according to the Scatchard equation,the characteristics of the complex between Sm(Ⅲ)-CAI and DNA belongs to types of intercalation of middle strength and partial electrostatic.
