RPHPLC Determination of Maslinic Acid in Spanish Oil Olive
摘 要
以枇杷叶为原料采用高效液相制备色谱技术制备了纯度达98.91%的山楂酸对照品,并用作制备标准曲线.油橄榄样品用乙醇-水(80+20)溶液90 ℃索氏回流提取24 h,在Kromasail C18色谱柱(250 mm×4.6 mm,5 μm)上分离,以甲醇-1%(体积分数)乙酸(88+12)混合溶液为流动相,流量1.0 mL·min-1,在215 nm波长处进行测定.此法用于油橄榄样品中山楂酸含量的检测,并在实样基础上加入山楂酸标准作方法的回收及精密度试验,测得平均回收率为98%,相对标准偏差(n=5)为1.21%.
Abstract
The oil olive sample was extracted with C2H5OH-H2O (80+20 by vol.) in a Soxhlet′s extractor at 90 ℃ for 24 h.An aliquot of the extract was used for separation on Kromasail C18 column,and a mixture of methanol and acetic acid solution (1+99) mixed in the ratio of 88 to 12 by volume was used as mobile phase flowing through the column at a rate of 1.0 mL·min-1.UV-detection at the wavelength of 215 nm was used in the determination.Reference material of maslinic acid was prepared from leaves of Eriobotrya japonica by the HPLC preparation system,giving purity of the product attained to 98.91%,which was used as the exteranal standard.Recovery and precision of the method were tested by addition of standard maslinic acid (20.0 mg) separately to 5 samples of oil olive and analyzed by the method given,value of average recovery found was 98%,and value of RSD (n=5) found was 1.21%.
