Resolution of Enantiomers of Chlorphenesin by HPLC with Mobile Phase Added with Chiral Additive
摘 要
以羟丙基-β-环糊精(HP-β-CD)作为流动相手性添加剂,建立了拆分氯苯甘醚对映体的高效液相色谱方法.测定中用ODS柱(4.6 mm×200 mm,5 μm)作为分离柱,用甲醇、乙腈、水体积比为25比25比50的混合液(其中含HP-β-CD 28.3 g·L-1)作为流动相,流量为0.5 mL·min-1,紫外检测波长为228 nm.按此方法氯苯甘醚对映体的分离度可达1.54.5次连续测定所得结果显示对映体的保留时间值的相对标准偏差分别为0.36%和0.11%;其峰面积值的相对标准偏差分别为0.83%和0.84%.
Abstract
Hydroxypropyl-β-cyclodextrin (HP-β-CD) was used as a chiral additive in the resolution of enantiomers of chlorphenesin by HPLC.The ODS Hypersil column (4.6 mm×200 mm,5 μm) was used as the stationary phase,and a mixture of CH3OH,acetonitrile and water (mixed in the ratio of 25∶25∶50 by vol.) containing 28.3 g·L-1 of HP-β-CD was used as mobile phase,with a flow-rate of 0.5 mL·min-1.UV-detection at the wavelength of 228 nm was adopted in the determination.The separation factor of this method attained to a value of 1.54.Values of RSD′s of retention time and of peak area of the enantiomers found in 5 repetitive determinations were 0.36%,0.11% and 0.83%,0.84% respectively.
