Separation and Determination of Sex Hormones by Mixed Micellar Electrokinetic Capillary Chromatography
摘 要
采用混合胶束-电动毛细管色谱法对性激素进行了分离测定,电泳介质为30 mmol·L-1十二烷基硫酸钠、40 mmol·L-1月桂酸、110 mmol·L-1氨水、异丙醇-水(15+85)溶液(pH=9.0),分离电压为20 kV,检测波长为214 nm.在优化的试验条件下,9种性激素均在13.5 min内完全分开,方法的检出限(3S/N)在0.2~2.7 mg·L-1之间.应用此方法分析了鹿鞭样品,测得回收率在85.0%~109.0%之间,测定值的相对标准偏差(n=6)在2.5%~5.5%之间.
Abstract
Mixed micellar electrokinetic capillary chromatography was applied to the separation and determination of sex hormones.A mixed solution containing 30 mmol·L-1 SDS,40 mmol·L-1 lauric acid,110 mmol·L-1 ammonium hydroxide and 2-propanol-water (15+85) solution was used as the electrophoretic medium.Separation voltage applied was 20 kV and wavelength of detection was taken at 214 nm.Under the optimum conditions,9 sex hormoneds was completed within 13.5 min.Values of detection limit (3S/N) of the method found were in the range of 0.2 to 2.7 mg·L-1.The proposed method was used in the analysis of sample of Penis et Testis Cervi,giving values of recovery and RSD′s (n=6) in the ranges of 85.0%-109.0% and 2.5%-5.5% respectively.
