Rapid Screening of Tenoxicam in Anti-Rheumatic Chinese Patent Drugs by IR in Combination with Discriminant Analysis

以30种市售抗风湿类中成药作为试验样品,并从中选取不含替诺昔康的15种样品作为阴性样品,每种阴性样品各取0.2 g左右,分别用甲醇10 mL超声提取,提取液经过滤后即得15种阴性样品溶液。取上述阴性样品15种,每种0.2 g左右,3份,依次加入替诺昔康10,15,20 mg,按上述方法用甲醇10 mL提取,制得45种阳性样品溶液。称取30种试验样品,每种0.2 g左右,按上述方法用甲醇10 mL提取,制得30种待测样品溶液。分别移取上述样品溶液各5 μL,采集其红外光谱图。用TQ Analyst 9.0软件及所得红外光谱图,按判别分析法建立模型,可对抗风湿类中成药中是否添加替诺昔康进行快速筛查。结果表明:所建模型的误判数为0,正确识别率为100%。
Abstract
Thirty kinds of anti-rheumatic Chinese patent drugs sold on market were taken as testing samples, and 15 of which, proved to contain no tenoxicam, were selected as the negative samples. Portions of about 0.2 g were taken from each of the negative samples, and were extracted ultrasonically with 10 mL of methanol separately. After filtering, the solution of the 15 negative samples were thus obtained. Three portions (about 0.2 g each) of each of the 15 negative samples were taken and 10, 15, 20 mg of tenoxicam were added respectively. These samples were extracted each with 10 mL methanol as described for the negative samples. Thus the solution of 45 positive samples were obtained. About 0.2 g sample portion was then taken from each of the 30 testing samples and extracted with 10 mL of methanol as described for the negative samples. Thus the solution from the 30 testing samples were obtained. Aliquots of 5 μL were taken from each of the above described solution and their IR spectra were taken. By using the soft ware TQ Analyst 9.0 and based on the IR spectra, discriminant analysis was applied to obtain the model for rapid screening of tenoxicam admixed into the anti-rheumatic Chinese patent drugs. It was found that by using the established model, rate of correct recognition of 100% was attained, with misdeeming case of zero.
