Rapid Determination of Acetaminophen and Pseudoephedrine Hydrochloride in Cold Drugs by UV Spectroscopy in Combination with the Algorithm of Vector Deduction
摘 要
为建立快速测定感冒药物中2种有效成分对乙酰氨基酚(AAP)和盐酸伪麻黄碱(PEPD)的数学模型,分别采集上述2种化合物的多波长紫外光谱,从而获得两者的标准光谱库(v1和v2)。同时取市售感冒药4种,各取1片,分别配成溶液后,各自采集其光谱-色谱联用数据并转化成各时刻全波长下的吸光度A。从中扣除与被测组分具有相同保留时间的数据阵列,得到不含有被测组分的数据矩阵,然后分别采用主成分分析和奇异值分解降维处理后,得到不含有AAP的本底数据库N1和不含有PEPD的本底数据库N2。分析样品时,取上述4种感冒药各1片,分别配制成溶液后,每一样品取2份,一份采集多波长紫外光谱数据,另一份进行高效液相色谱(HPLC)分析。从而建立样品的光谱库a,并运用向量扣减方法计算得到样品中2种有效成分的含量。AAP测定值的相对标准偏差(n=5)均小于2.0%,PEPD测定值的相对标准偏差(n=5)均小于4.0%;加标回收试验测得回收率分别为94.5%~106%和92.1%~108%。此方法的测定结果与HPLC的测定结果之间的相对误差分别在±4.0%和±3.0%以内。
Abstract
To establish a mathematical model for rapid determination of 2 effective constituents in cold drugs, i.e., acetaminophen (AAP) and pseudoephedrine hydrochloride (PEPD), multiwavelength UV spectra of the 2 compounds were collected respectively, from which database of standard spectra (v1 and v2) of these 2 compounds were obtained. And at the same time, multiwavelength chromatograms of 4 cold drugs obtained from the market were collected separately by UV spectroscopy and HPLC in hyphenation in solutions prepared by dissolving 1 piece each of the 4 drug tablets, and data of the multiwavelength chromatograms were then transformed into values of absorbances (A) under whole wavelength at various moments. Values of absorbances of those substances having their retention times same as the analyte were subtracted from the values of A, and the data matrix not involving the constituent to be determined was obtained. And thence, after chemometric treatment by principle component analysis and resolution of unusual values, matrix database of N1 not containing AAP, and also the matrix database of N2 not containing PEPD were obtained. In the analysis of unknown samples of the drugs, one piece of each of the drugs was taken and dissolved as prescribed. Two aliquots were taken from each sample solution, one for collecting the UV spectroscopic data and the other for HPLC analysis. Spectra database a of the sample was established based on the data obtained. Results of contents of AAP and PEPD were found by using the algorithm of vector deduction. Values of RSDs (n=5) found for AAP were less than 2.0%, and for PEPD were less than 4.0%. Values of recovery found by standard addition method were in the ranges of 94.5% to 106% and 92.1% to 108% respectively. Values of the results of AAP and PEPD found by the present method were checked by the HPLC method, relative deviations between values of the 2 constituents determined by the 2 methods were within ±4.0% and ±3.0% respectively.
