Determination of Patulin in Canned Supplementary Food Fruit Puree for Infants and Young Children by Ultra-High Performance Liquid Chromatography Tandem Mass Spectrometry with Solid Phase Extraction

提出了固相萃取-超高效液相色谱-串联质谱法测定婴幼儿罐装辅助食品果泥中展青霉素含量的方法。果泥样品经果胶酶酶解、乙酸乙酯提取后,经MAX固相萃取柱净化,浓缩复溶后采用Waters BEH C18色谱柱分离,电喷雾离子源负离子和多反应监测模式下进行测定,同位素内标法定量。结果表明,展青霉素标准曲线的线性范围为10.0~200 μg·L-1,检出限(3S/N)为2 μg·kg-1。按标准加入法进行回收试验,回收率为101%~112%,测定值的相对标准偏差(n=6)均小于4.0%。该方法应用于33份市售的婴幼儿罐装辅助食品果泥样品中展青霉素的测定,均未检出。
Abstract
A method was proposed for the determination of patulin in canned supplementary food fruit puree for infants and young children by ultra-high performance liquid chromatography tandem mass spectrometry with solid phase extraction. The sample was hydrolyzed by pectinase, extracted by ethyl acetate, purified by MAX solid phase extraction column. The separation was performed on Waters BEH C18 column, then the detection was performed in the electrospray ion source negative ion and multiple reaction monitoring modes. The isotope internal standard method was used for quantification. As shown by the results, linear range of standard curve for patulin was 10.0-200 μg·L-1, with detection limit (3S/N) of 2 μg·kg-1. Test for recovery was made by standard addition method, giving results in the range of 101%-112%, with RSDs (n=6) of the determined values less than 4.0%. This method was applied to determination of patulin in 33 commercially available canned supplementary food fruit puree samples, and patulin was not detected.
