Application of Characteristic Fingerprints Combined with Pattern Recognition Method in Quality Control of Lobelia Chinensis Lour
摘 要
采用高效液相色谱法对全国各产地的半边莲药材样品和半边莲饮片样品进行分析,建立半边莲的特征图谱,确定半边莲有5个共有特征峰,1号峰和3号峰分别为香叶木苷和蒙花苷,可将蒙花苷峰作为参照物峰。成功地采用正交偏最小二乘法-判别分析对特征图谱进行了拟合建模,香叶木苷和蒙花苷对半边莲药材和半边莲饮片的分类贡献较大。香叶木苷和蒙花苷的质量在一定范围内与其对应的色谱峰面积呈线性关系,检出限(3S/N)依次为0.030 4,0.031 8 mg·L-1。香叶木苷和蒙花苷的加标回收率依次为97.7%,100%,测定值的相对标准偏差(n=6)依次为1.3%,2.5%。方法用于半边莲药材样品和半边莲饮片样品的分析,半边莲药材中香叶木苷和蒙花苷的含量均高于半边莲饮片中香叶木苷和蒙花苷的含量。
Abstract
Samples of medicinal materials and decoction pieces of Lobelia Chinensis Lour from various regions in China were analyzed by HPLC to establish characteristic fingerprints of Lobelia Chinensis Lour, and 5 common peaks were identified as characteristic peaks of Lobelia Chinensis Lour. Among them, peak 1 was diosmin and peak 3 was linarin. The peak of linarin could be used as the reference peak. The characteristic fingerprints were fitted and modeled by orthogonal partial least squares-discriminant analysis,and the model was made successfully. Diosmin and linarin were found to be great contributions to the classification of medicinal materials and decoction pieces of Lobelia Chinensis Lour. Linear relationships between values of chromatographic peak area and mass of diosmin and linarin were kept in definite ranges, with detection limits (3S/N) of 0.030 4, 0.031 8 mg·L-1 respectively. Values of recovery of diosmin and linarin found by standard addition method were 97.7%, 100% respectively, and values of RSDs (n=6) of determined values were 1.3%, 2.5% respectively. The proposed method was applied to the analysis of samples of medicinal materials and decoction pieces of Lobelia Chinensis Lour. The contents of diosmin and linarin in medicinal materials were higher than those in decoction pieces.
